So when a compost is 100% full (50 fill points).
Does it generate 1 humus per 1 fill point ore does it take 2 fill points to generate 1 humus?

It takes 1 fill point per 1 humus so you will see the % full drop by 2 each time.

How many pumpkins does one 100% field have?

It actually depends on the growth stage, it's 5 for stage 1 and less for higher (2 for stage 3 if I'm not mistaken). That way you always get the same amount of seeds 6x5=2x15, it's probably true for stage 2 as well. Personally I've never grown stage 2 pumpkins, since they're pretty useless.
