I have started venturing into the darkness further and further, i am wondering how do i cope with rats and disease? is there a way i can one hit kill rats from far away? also darkness creatures what is the best way to kill them?

I am experienced and can kill them with punch before they attack also halberd or just go witchy

Punch is for amateur, halberd is good or if you don't have the skill use roundhouse kick.
