I've heard the dirt just regenerates to whatever biome is next to it. You can't start a new one.

While your first statement is true, Mereni, deforested lands to turn to grass. There are quite a few interesting terraforming tricks in HnH, and I'll be surprised if they don't eventually show up here.

Does anyone know what the minimum size of cleared land is to get grassland?

There is no real minimum.

Best thing to do, is just level a large amount of land, and put a pavement border around it, so the original biome cannot grow back inside. After a couple of weeks / months, the area inside will turn into grassland. If you then want to expand the grassland, level more land outside the pavement you made, then remove the pavement so the grassland can expand.

If you do not make a pavement border, the original biome will start to grow back, and make progression / development of your grass land slower as you will need to keep turning over the biome dirt so the grass land can expand.

Note : you do not need to level land perfectly, just merely turn it over to dirt.

Last time I paved I had cleared roughly 10x15 squares but the biome grew back over the outer paved area. Not sure it would've grown over it all if I had left it. I'll try your suggestion.

So with this being said if you cut down a tree and do not remove the stump will a new tree eventually grow?

No, the stump will turn into an old stump just like the ones you find all over the world.
