Which client are you using? With the default client, there is no external config file. Since the game is loaded using Java Web Start, things can get messy with the cache and sometimes settings will get saved into the Registry in Windows. With Ender's, some of the config information is saved in the user folder (<user>/Salem in Windows if I recall correctly).
Open the options window (Ctrl+O or the options button in the client). There should be sound settings you can adjust there. Make sure the sound is set to on. If it was already on, turn it off, restart, and then turn them on to see if you get sounds again.
Was anything else on your computer changed that you are aware of at about that time? Things such as Java and driver updates can mess with these things. If Java itself is the problem, you should uninstall Java, use a tool such as CCleaner to clear out any remaining files and Registry entries (assuming Windows user), and then reinstall.
